Ride back out the road that you drove in on to the spring (248) for two miles. The bike path is on the right just before you get back to (249/129). The bike path from there only goes south to Branford, so you can't go wrong. Just follow the bike path all the way to Ft. White or Oleno.
In fact you will probably notice the bike path as you drive up 249/129 to the spring because it paralels 249/129, on the west side of the road.
 
Incidentally, there is a small error caused by the difficulty of reading some numbers across map lines on the computer, on the route recommended for the trip to Little River. The road that goes west from 249/129 is 248, not 240. The description below is corrected..  My apologies for the error.
 
To get to Little River Spring from Gainesville:
From the Intersection of NW 39th and I-75 proceed north on I-75for 9 miles and take the exit to High Springs. Proceed to High Springs on 441, take US 27 out of High Springs to Ft. White and Branford, turn north at the light in Branford on 249/129 and proceed  for about 3 miles to CR 248. You should see a sign pointing to Little River Springs at this point. Proceed about another 2 miles west to the spring on CR 248 Total distance, about 45 miles.
